چکیده

Soluble proteins make up 6.4–8.3% of plasma by weight [1], a value comparable to their concentration in cell water [2]. Many plasma proteins are synthesized by the liver. Their concentrations may decrease in the setting of liver disease, malnutrition or protein-losing states such as nephrotic syndrome, severe burns, bowel inflammation, intestinal lymphangiectasia or major bleeding episodes. The most abundant plasma proteins have little or no enzymatic activity. Instead, their function is typically binding to other molecules, generically referred to as ligands. Some plasma binding proteins that have been characterized are listed in Table 1. Most of these proteins bind to only one (or a few related) molecules with high affinity. For example, haemopexin binds haem, retinol-binding protein binds retinol, and transferrin binds iron. In contrast, other binding proteins such as albumin and alpha-fetoprotein are far less selective. Albumin is the most important serum binding protein in terms of concentration (about 4.2%) [1,3] and number of different ligands bound. The latter include fatty acids [4], unconjugated bilirubin [5], many divalent cations such as copper [6,7], hydrophobic bile acids [6] and a variety of drugs and toxins [6,8–11] (Table 2). Albumin’s broad specificity reflects not only 2.4 Synthetic function

Relationship between serum levels of chromium and iron with transferrin and albumin levels as carrier proteins in diabetic patients with atherosclerosis

Backgrounds: Type 2 diabetes mellitus is one of the most common metabolic diseases in the world, resulting in complete or partial deficiency or resistance to insulin action. In this study, the association between serum levels of chromium and iron with serum levels of their carriers, transferrin and albumin in diabetic patients with atherosclerosis was investigated. Materials and methods: In ...

A novel approach for measuring sphingosine-1-phosphate and lysophosphatidic acid binding to carrier proteins using monoclonal antibodies and the Kinetic Exclusion Assay.

Sphingosine-1-phosphate (S1P) and lysophosphatidic acid (LPA) are bioactive signaling lysophospholipids that activate specific G protein-coupled receptors on the cell surface triggering numerous biological events. In circulation, S1P and LPA associate with specific carrier proteins or chaperones; serum albumin binds both S1P and LPA while HDL shuttles S1P via interactions with apoM. We used a s...

Early plasma protein and mineral changes after surgery: a two stage process.

Sequential changes in albumin, transferrin, alpha 1-acid glycoprotein, C reactive protein, fibrinogen, copper, iron, and zinc in plasma up to 24 h after hysterectomy were measured. No increases in the concentrations of the acute phase proteins alpha 1-acid glycoprotein, C reactive protein, and fibrinogen were observed until 6 h after the skin incision. These increases were preceded by significa...

Antagonism of Insulin Action on Muscle by the Albumin-bound B Chain of Insulin.

1. The presence of a substance associated with human albumin that exerts anti-insulin activity on the isolated rat diaphragm has been confirmed. This factor has been removed from albumin, thereby providing a source of non-antagonistic carrier protein. 2. Derivatives of the polypeptide B chain of insulin obtained by chemical scission of the interchain disulphide bonds have been separated by conv...

Interaction with Serum Albumin As a Factor of the Photodynamic Efficacy of Novel Bacteriopurpurinimide Derivatives

Optimization of the chemical structure of antitumor photosensitizers (PSs) is aimed at increasing their affinity to a transport protein, albumin and irreversible light-induced tumor cell damage. Bacteriopurpurinimide derivatives are promising PSs thanks to their ability to absorb light in the near infrared spectral region. Using spectrophotometry, we show that two new bacteriopurpurinimide deri...
